rTBL-1 is a recombinant lectin derived from the Tepary bean (Phaseolus acutifolius) with demonstrated cytotoxic activity against colon cancer cell lines. Its mechanism involves recognition of β1-6 branched N-glycans, which are overrepresented on several tumor cell types, leading to apoptosis of cancer cells. The interaction with these glycans is mediated by specific arginine residues (R103, R130) in the carbohydrate binding pocket, and mutational studies confirm their importance for activity. rTBL-1's cytotoxic effect is largely independent of epidermal growth factor receptor (EGFR) pathways, although EGFR-related N-glycans may contribute to more pronounced cytotoxic responses. The protein is reported to partially resist gastrointestinal digestion and can interact with the intestinal membrane, which may be relevant for development as an oral or other administrable agent. Its mechanism partly involves p38 MAPK pathway signaling. rTBL-1 is being studied mainly as a potential anticancer agent, with most investigations focused on colorectal cancer cells.[1][3][4][5][7][8][10][14]
