Overview

RTL1000 is a recombinant T-cell receptor ligand (RTL) designed as a single-chain protein construct containing the outer two domains of HLA-DR2 linked to the myelin oligodendrocyte glycoprotein (MOG) 35–55 peptide. It was developed as an immunomodulatory and neuroprotective therapy for multiple sclerosis (MS), aiming to induce antigen-specific immune tolerance without broad immunosuppression and potentially promote neural repair. Preclinical studies demonstrated that analogues of RTL1000 could reverse disease in animal models of MS, while early clinical trials showed it was generally well tolerated at doses up to 60 mg. The mechanism involves modulation of T cell function, inhibition of pathogenic immune responses, and reduction in inflammatory infiltration into the central nervous system. There is also evidence from preclinical stroke models that RTL1000 can reduce infarct size and improve functional recovery by limiting neuroinflammation[1][5][6].
