RTP004 is a proprietary 35-amino acid, highly positively charged, lysine- and arginine-rich bi-cell-penetrating peptide (bi-CPP) developed by Revance Therapeutics. It serves as a novel stabilizing excipient in the formulation of daxibotulinumtoxinA (brand name DAXXIFY). RTP004 binds noncovalently to the 150 kDa botulinum toxin type A (BoNT/A) molecule, replacing the need for human serum albumin. Its primary functions include providing formulation stability by preventing self-aggregation and surface adsorption, enhancing the binding of the neurotoxin to neuronal cell membranes, and facilitating the internalization of the toxin into cells. Additionally, it helps reduce post-injection diffusion, potentially contributing to the long duration of effect observed with daxibotulinumtoxinA. This represents the first CPP-containing product approved for clinical use.
