The rTSST-1 variant vaccine is a recombinant protein-based vaccine designed to prevent toxic shock syndrome caused by Staphylococcus aureus. It contains a detoxified double-mutant form of the toxic shock syndrome toxin 1 (TSST-1), which is a superantigen responsible for triggering life-threatening immune responses in humans. The mechanism of action involves inducing anti-toxic immunity by stimulating the production of neutralizing antibodies against TSST-1, thereby preventing or mitigating the effects of this superantigen in case of infection. The vaccine has been shown in phase 2 clinical trials to be safe, well-tolerated, and highly immunogenic in healthy adults aged 18–64 years. Two doses of 100 μg provided persistent immune responses lasting up to at least 18 months post-vaccination[2][3][9]. Developed as part of efforts to address an unmet medical need for specific prevention against staphylococcal toxic shock syndrome, it may also be used for hyperimmunization protocols for immunoglobulin production[6].
