Overview

RTX-1688 is a first-in-class, orally available small-molecule inhibitor of 15‑prostaglandin dehydrogenase (15‑PGDH; hydroxyprostaglandin dehydrogenase), originally developed by Rodeo Therapeutics and later acquired by Amgen, for tissue regeneration and repair in inflammatory diseases, with inflammatory bowel disease (IBD) as the lead indication.[1][2][7][11][13] By inhibiting 15‑PGDH, RTX-1688 elevates tissue levels of prostaglandin E2 (PGE2), a key mediator of epithelial barrier repair and stem cell self-renewal, thereby promoting mucosal healing and regeneration in preclinical models of injury-induced IBD and potentially other conditions where tissue repair is crucial.[1][2][7][10][13] Extensive preclinical studies have shown marked monotherapy efficacy in IBD models and favorable in vitro and in vivo characteristics, and the compound progressed through IND-enabling, preclinical development before the program was later reported as discontinued at the preclinical stage.[1][2][3][7][11]
