Overview

RTX-212 is an allogeneic red cell therapeutic developed by Rubius Therapeutics using its proprietary Red Cell Therapeutic (RCT) platform. It consists of red blood cells genetically engineered to co-express 4-1BB ligand (4-1BBL) and an IL-15/IL-15Rα fusion protein (IL-15TP) on their surface. This dual-ligand approach is designed to provide coordinated activation of co-stimulatory (4-1BB) and cytokine (IL-15) pathways, promoting the proliferation, survival, and cytotoxic activity of CD8+ T cells and natural killer (NK) cells. By presenting these ligands in their native form through cell-cell contact and restricting their distribution to the vascular system, RTX-212 aims to enhance anti-tumor efficacy while minimizing the systemic toxicities often associated with recombinant agonist antibodies and cytokines. Preclinical studies demonstrated potent in vitro and in vivo activity in melanoma and colorectal cancer models, showing significant tumor growth inhibition and increased immune cell infiltration.
