Overview

RTX-224 is an allogeneic, off-the-shelf cellular therapy developed by Rubius Therapeutics using its proprietary RED PLATFORM. It consists of engineered red blood cells that express hundreds of thousands of copies of 4-1BB ligand (4-1BBL) and interleukin-12 (IL-12) on their surface. The drug is designed as a broad immune agonist, activating both adaptive and innate immune responses by stimulating CD8+ and CD4+ T cells, promoting antigen presentation, and activating natural killer (NK) cells. This mechanism aims to produce a potent anti-tumor response in solid tumors with known sensitivity to T cell killing, such as those with high mutational burden or PD-L1 expression. Clinical development was focused on advanced solid tumors including non-small cell lung cancer, melanoma, head and neck squamous cell carcinoma, urothelial carcinoma (bladder), triple-negative breast cancer, cutaneous squamous cell carcinoma, transitional cell carcinoma, and other late-stage or refractory solid tumors[1][2][3][4][5][7].
