Ru360 (ruthenium-360) is a cell-permeable, oxygen-bridged dinuclear ruthenium ammine complex that acts as a highly selective inhibitor of the mitochondrial calcium uniporter (MCU). It binds to mitochondria with high affinity and potently blocks Ca2+ uptake into mitochondria in vitro and in situ. Ru360 does not significantly affect other cellular calcium transport processes at micromolar concentrations. Its mechanism of action involves preventing mitochondrial calcium overload by inhibiting MCU-mediated Ca2+ entry, thereby protecting against conditions such as myocardial reperfusion injury, liver steatosis induced by high-fat diet, and postoperative cognitive dysfunction. Ru360 has been studied for its protective effects on cardiac tissue during ischemia-reperfusion injury and for neuroprotection in models of brain injury or cognitive impairment[1][3][6][8][10].
