Rucaparib is a small molecule, orally administered poly (ADP-ribose) polymerase (PARP) inhibitor used as an antineoplastic agent. It selectively inhibits PARP enzymes, particularly PARP-1, PARP-2, and PARP-3, which are involved in DNA repair. By blocking these enzymes, rucaparib exploits the concept of synthetic lethality in cancer cells with deficiencies in homologous recombination repair genes such as BRCA1 and BRCA2. This leads to accumulation of DNA damage and subsequent cancer cell death. Rucaparib is primarily indicated for the maintenance treatment of adults with recurrent epithelial ovarian, fallopian tube, or primary peritoneal cancer who are in a complete or partial response to platinum-based chemotherapy. It is also approved for certain patients with metastatic castration-resistant prostate cancer harboring deleterious BRCA mutations who have received prior therapies[1][3][4][9].
