RUNX1 mRNA is a preclinical mRNA-based therapeutic being developed by NANO mRNA (now Nano Holdings, Inc.) for the treatment of knee osteoarthritis. The drug utilizes a polyplex nanomicelle delivery system composed of polyethylene glycol (PEG)-polyamino acid block copolymers to deliver mRNA encoding the transcription factor RUNX1 (runt-related transcription factor 1) directly to articular cartilage. By restoring or augmenting RUNX1 expression, the therapy aims to promote chondrocyte proliferation, enhance cartilage-anabolic markers, and suppress the progression of osteoarthritis, potentially providing both structural repair and pain reduction.
