Overview

Rupintrivir is a synthetic small molecule peptidomimetic inhibitor developed as an antiviral agent targeting human rhinovirus (HRV) infections. It acts as an irreversible inhibitor of the viral 3C protease, a key enzyme required for processing viral polyproteins and thus essential for viral replication. By inhibiting this protease, rupintrivir blocks the maturation of viral proteins and prevents virus replication in host cells. The drug has demonstrated broad-spectrum activity against all tested HRV serotypes *in vitro* and also shows inhibitory effects against enteroviruses and noroviruses by targeting their respective 3C or 3C-like proteases. Rupintrivir was primarily developed for intranasal administration to treat or prevent the common cold caused by rhinovirus infection, but development was discontinued after Phase II trials due to lack of sufficient efficacy in natural infection studies[1][2][3][4][5][6].
