Overview

Rusvinoxidase is an L-amino acid oxidase (LAAO) purified from the venom of the Russell's viper (Daboia russelii russelii).[1][2][4] It is an acidic monomeric glycoprotein (~57 kDa) that induces apoptosis in certain human cancer cell lines (e.g., MCF-7 breast cancer and Colo-205 colorectal cancer) but shows differential cytotoxicity depending on the cellular environment.[1][2][4] Rusvinoxidase promotes apoptosis predominantly via the extrinsic (death-receptor, caspase-8-mediated) and intrinsic (mitochondrial, ROS-dependent) pathways, leading to ROS generation, glutathione depletion, catalase activity reduction, mitochondrial depolarization, chromatin condensation, and activation of effector caspases.[1][2][4][9] It downregulates anti-apoptotic proteins (Bcl-xL, HSP-90, HSP-70) and increases pro-apoptotic proteins (Bax), causing cytochrome c release and caspase activation.[4][9] Rusvinoxidase is non-toxic in mice at tested doses, suggesting potential for anticancer drug development.[4][13]
