Ruxoprubart (NM8074) is a humanized monoclonal antibody that selectively binds to the Bb fragment of complement factor B. By targeting Bb, it blocks the formation and activity of the alternative pathway C3 convertase (C3bBb), thereby inhibiting activation of the alternative complement pathway while preserving the classical pathway. This selectivity allows for effective control of complement-mediated diseases without compromising host defense against infections. Ruxoprubart is being developed primarily for paroxysmal nocturnal hemoglobinuria (PNH), but also has potential applications in other renal and inflammatory disorders such as IgA nephropathy (IgAN), C3 glomerulopathy (C3G), atypical hemolytic uremic syndrome (aHUS), anti-neutrophil cytoplasmic antibody-associated vasculitis (AAV), membranoproliferative glomerulonephritis, and dermatomyositis. The drug has demonstrated a favorable safety profile in phase I trials with healthy volunteers and met all clinical endpoints in phase II PNH studies[2][6][7][8][9].
