RV-1088 is an inhaled, small-molecule narrow spectrum kinase inhibitor originally developed by RespiVert to treat severe inflammatory airway diseases such as asthma and chronic obstructive pulmonary disease. It is designed to simultaneously inhibit a defined subset of pro‑inflammatory kinases including p38 MAPKs (notably p38α and p38γ), Src family kinases (such as c-Src and Hck) and spleen tyrosine kinase (Syk), thereby suppressing downstream signaling pathways that drive production of key inflammatory cytokines including TNF-α, IL‑6 and IL‑8 in macrophages, monocytes and synovial fibroblasts.[7][8][4][15][17] Preclinical studies have shown that RV-1088 potently reduces cytokine release with low nanomolar IC50 values and is more effective than single kinase inhibitors or their combinations, supporting the concept that coordinated inhibition of multiple signaling cascades can provide superior anti‑inflammatory efficacy.[7][8][4]
