Overview

rV-NY-ESO-1 is a recombinant vaccinia virus-based cancer vaccine engineered to express the NY-ESO-1 antigen, a highly immunogenic cancer/testis antigen. The vaccine is designed to stimulate both humoral and cellular immune responses against tumors expressing NY-ESO-1 by presenting the antigen in the context of viral infection, thereby enhancing immunogenicity. It has been studied as part of prime/boost regimens (often with fowlpox-based rF-NY-ESO-1) in patients with advanced cancers such as melanoma and epithelial ovarian cancer. Clinical trials have demonstrated that vaccination with rV-NY-ESO-1 can induce robust CD4+ and CD8+ T-cell responses targeting multiple epitopes of NY‑ESO‑1, leading to clinical benefit including disease stabilization and objective tumor responses in some patients[6][8]. The approach leverages the restricted expression pattern of NY‑ESO‑1 (mainly in germ cells and various tumors) to minimize off-target effects while maximizing anti-tumor immunity.
