RV1729 is a clinical-stage, small molecule inhibitor targeting the phosphoinositide 3-kinase gamma and delta isoforms (PI3Kγ and PI3Kδ). It is being developed as an inhaled anti-inflammatory therapy for respiratory diseases, specifically asthma and chronic obstructive pulmonary disease (COPD). By inhibiting PI3Kγ and PI3Kδ, RV1729 reduces airway hyper-responsiveness, IgE release, pro-inflammatory cytokine expression, inflammatory cell accumulation in the lung, and vascular permeability. The drug was originally developed by RespiVert Ltd., later acquired by Janssen Biotech (a Johnson & Johnson company), with licensing to Pulmatrix in 2017. Clinical studies have focused on its safety, tolerability, pharmacokinetics, and pharmacodynamics in both healthy volunteers and patients with asthma or COPD[1][2][3][4][5][6].
