Overview

RV568 is a narrow-spectrum kinase inhibitor with selectivity for p38 mitogen-activated protein kinase (MAPK)-α and -γ isoforms, as well as SRC family kinases. It is being developed primarily for the treatment of chronic obstructive pulmonary disease (COPD) and has also been considered for asthma. RV568 exhibits potent anti-inflammatory effects in monocytes, macrophages, and bronchial epithelial cells—often greater than those observed with corticosteroids or other p38 inhibitors such as Birb796. In preclinical models, it demonstrated significant inhibition of cytokine release and neutrophilic airway inflammation. Inhaled RV568 was evaluated in a 14-day clinical trial in COPD patients, where it improved lung function (pre-bronchodilator FEV1) and reduced sputum malondialdehyde levels compared to placebo. The drug was well-tolerated with adverse events similar to placebo[1][2][4][5][6].
