Overview

RVB-001 is a genetically engineered probiotic developed by ResVita Bio for the treatment of Netherton syndrome, a rare and life-threatening genetic skin disorder. The therapy consists of non-pathogenic bacteria that are topically applied to the skin, where they temporarily colonize and continuously produce and release LEKTI (Lympho-epithelial Kazal-type-related inhibitor), a serine protease inhibitor encoded by the SPINK5 gene. In Netherton syndrome, mutations in SPINK5 lead to a deficiency of LEKTI, resulting in uncontrolled activity of kallikrein-related peptidases (KLKs) such as KLK5, KLK7, and KLK14. This excessive proteolysis causes premature exfoliation of the skin and severe barrier defects. By restoring LEKTI levels locally, RVB-001 aims to inhibit these proteases, reduce inflammation, and restore the integrity of the epidermal barrier. The FDA granted RVB-001 Rare Pediatric Disease Designation in 2022.
