RVL-CLR01

RVL-CLR01 (also known as REVTx-99) is a synthetic Toll-like receptor 4 (TLR4) agonist developed by Revelation Biosciences as an intranasal immunotherapy for the prevention and treatment of respiratory viral infections. The active ingredient is phosphorylated hexaacyl disaccharide (PHAD), a synthetic analog of monophosphorylated lipid A (MPLA). RVL-CLR01 is designed to stimulate the innate immune response within the upper respiratory tract, inducing the production of endogenous interferons (such as IFN-beta) and protective cytokines to create an antiviral state. This broad-spectrum approach was intended to address various viral pathogens, including SARS-CoV-2, influenza, and respiratory syncytial virus (RSV). Clinical development for viral indications was largely discontinued following a Phase 2b viral challenge study in influenza that failed to meet its primary endpoint.
