rVSV∆G-LASV-GPC is a recombinant, replication-competent vesicular stomatitis virus (rVSV) vector vaccine candidate engineered to express the glycoprotein precursor (GPC) of the Lassa virus (LASV), specifically from the Josiah strain. The vaccine lacks the native VSV G protein, which is replaced by LASV GPC, enabling it to induce immune responses against Lassa fever. Preclinical studies have demonstrated that a single dose provides robust protection in non-human primates and guinea pigs against lethal LASV challenge, inducing high titers of anti-LASV IgG and activating both CD4+ and CD8+ T cell responses. Clinical trials are ongoing in West Africa to assess safety, tolerability, and immunogenicity in adults and children. The vaccine is being developed as a single-dose regimen for rapid deployment during outbreaks or for routine immunization in endemic regions[1][5][9].
