rVSV-NDV is a chimeric oncolytic virus engineered as a hybrid vector for the treatment of solid tumors, particularly hepatocellular carcinoma (HCC). The virus utilizes a vesicular stomatitis virus (VSV) backbone where the native glycoprotein (G) has been replaced by the hemagglutinin-neuraminidase (HN) and a modified, hyperfusogenic fusion (F) protein from the Newcastle disease virus (NDV). This modification is designed to enhance safety by abrogating the neurotoxicity and hepatotoxicity associated with wild-type VSV while maintaining potent oncolytic activity. rVSV-NDV works by inducing tumor-specific syncytium formation, which facilitates rapid cell-to-cell spread and triggers immunogenic cell death. Preclinical studies have demonstrated that systemic administration of the virus leads to significant survival prolongation in immune-competent models of HCC.
