rVSV-UL141 is a recombinant oncolytic virus based on vesicular stomatitis virus (VSV) engineered to express the UL141 protein from human cytomegalovirus. UL141 acts by downregulating the NK cell-activating ligand CD155, thereby inhibiting the recruitment and function of natural killer (NK) and NKT cells within tumor tissue. This modification enhances intratumoral virus replication, leading to improved tumor necrosis and prolonged survival in preclinical models of hepatocellular carcinoma (HCC). The drug is being developed as an oncolytic virotherapy for cancer, specifically for multi-focal HCC, with future applicability to other cancers suggested[1][4].
