rVSVN4CT1-EBOVGP1 is a monovalent, live-attenuated recombinant viral vector vaccine designed to prevent Ebola virus disease caused by the Zaire ebolavirus. Developed using the VesiculoVax platform, the vaccine utilizes a recombinant vesicular stomatitis virus (rVSV) backbone engineered for enhanced safety through two primary modifications: the relocation of the nucleoprotein (N) gene to the fourth position (N4) and the truncation of the glycoprotein cytoplasmic tail to a single amino acid (CT1). These modifications attenuate the virus and reduce neurovirulence while maintaining its ability to induce robust humoral and cellular immune responses. The vaccine expresses the glycoprotein (GP) of the Zaire ebolavirus (Mayinga strain). Originally developed by Profectus BioSciences with significant funding from BARDA, the DoD, and NIAID, the program was later acquired by Auro Vaccines (a subsidiary of Aurobindo Pharma). It has demonstrated 100% protection in non-human primate models and has been evaluated in Phase 1 clinical trials.
