RVT-103

RVT-103 is an oral fixed-dose combination of the acetylcholinesterase inhibitor donepezil and the peripherally acting antimuscarinic agent glycopyrrolate, developed by Axovant Sciences (later Sio Gene Therapies) to treat dementia, particularly Alzheimer’s disease and related disorders.[1][5] The rationale is to maintain or enhance central cholinergic signaling via donepezil to improve cognition, while using glycopyrrolate to mitigate dose-limiting peripheral cholinergic adverse effects such as gastrointestinal symptoms by blocking muscarinic receptors outside the central nervous system.[1][8] The program was part of Axovant’s dementia pipeline alongside intepirdine and nelotanserin, but there is no evidence that RVT-103 advanced beyond early-stage development or reached regulatory approval.[5][7]
