RVU-27065 is a preclinical, third-generation small-molecule stimulator of interferon genes (STING) agonist developed by Ryvu Therapeutics for oncology and potentially immunotherapy applications. It is a non-nucleotide small molecule designed to activate all major human STING haplotypes, induce robust type I interferon and proinflammatory cytokine signaling, and generate antitumor immune responses, with preclinical models showing dose-dependent tumor regression and complete responses in syngeneic breast cancer models.[1][3][5][7][9] The compound is intended both as a standalone systemic therapy and as a targeted delivery payload (for example, in conjugated formats such as ADCs), and has shown favorable drug-like properties and a good safety profile in nonclinical studies, leading to a licensing and collaboration deal in which BioNTech obtained rights to Ryvu’s STING agonist portfolio including RVU-27065.[1][3][6][8][9]
