RVx-1001

RVx-1001 is a live-attenuated herpes simplex virus type 1 (HSV-1) prophylactic vaccine candidate being developed by Rational Vaccines. Also known as VC2 or HSV-1(VC2), the vaccine is designed to prevent genital herpes, herpetic keratitis, and other HSV-1-related diseases. The vaccine is engineered using genetic modifications—specifically deletions in the amino termini of glycoprotein K (gK) and the UL20 protein, or mutations in the ICP0 gene—to attenuate viral virulence. These modifications prevent the virus from entering neuronal axons and establishing latency while allowing it to replicate sufficiently to stimulate a robust immune response. Preclinical studies have demonstrated that RVx-1001 provides protection against lethal ocular and genital HSV-1 challenges.
