RVx201 is a live attenuated, rationally engineered therapeutic vaccine candidate developed for the treatment of herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2) infections. The vaccine is designed to be interferon-sensitive by mutating a specific viral protein that normally allows HSV to evade the host immune response, thereby enhancing immunogenicity and reducing the virus's ability to inhibit immune defenses. Preclinical studies have shown that RVx201 significantly reduces recurrent genital lesions and symptomatic days in animal models compared to other vaccines or placebo. It elicits robust cellular immune responses against HSV infection. The vaccine was originally created at Southern Illinois University School of Medicine and the University of Kansas, with development led by Rational Vaccines[1][3][6].
