RVx202 is a novel live-attenuated herpes simplex virus type 2 (HSV-2) vaccine candidate developed by Rational Vaccines, Inc. for the prevention and treatment of genital herpes. The vaccine is designed to elicit a robust immune response while avoiding the establishment of latency in the dorsal root or sacral sympathetic ganglia, a key feature of wild-type HSV-2 infection. Preclinical studies in guinea pig models have demonstrated safety and efficacy, with no development of HSV-2-associated disease following vaginal or intradermal inoculation. The vaccine candidate has shown transient viral shedding in vaginal secretions for three days post-inoculation in animal models, indicating active but controlled replication.
