RWQWRWQWR-Ahx-RRRRRRRR is a synthetic hybrid peptide designed for anticancer activity, specifically targeting cervical cancer. It consists of a palindromic core sequence (RWQWRWQWR) derived from bovine lactoferricin (LfcinB) linked via a 6-aminohexanoic acid (Ahx) spacer to an octa-arginine (RRRRRRRR) cell-penetrating peptide (CPP). The mechanism of action involves initial electrostatic interactions between the positively charged arginine residues and the negatively charged components of the cancer cell surface, followed by membrane disruption or internalization facilitated by the hydrophobic tryptophan residues. In vitro studies demonstrate significant cytotoxic activity against HeLa and Ca Ski cervical cancer cell lines, with a rapid effect observed within 2 hours. The peptide was developed by researchers at the Universidad Nacional de Colombia and Pontificia Universidad Javeriana as part of a strategy to enhance the selective cytotoxic effect of bioactive peptides against tumor cells.
