RWQWRWQWR-Ahx-YGRKKRPQRRR is a synthetic hybrid peptide designed for anticancer activity, specifically targeting cervical cancer. It consists of a core palindromic sequence (RWQWRWQWR) derived from bovine lactoferricin (LfcinB) linked via a 6-aminohexanoic acid (Ahx) spacer to a cell-penetrating peptide (CPP) sequence (YGRKKRPQRRR). The peptide's mechanism of action involves an initial electrostatic interaction between its positively charged arginine residues and the negatively charged components of the cancer cell surface, followed by membrane disruption or internalization facilitated by tryptophan residues and the CPP fragment. In vitro studies have demonstrated that this hybrid peptide exhibits significant, rapid, and concentration-dependent cytotoxic activity against HeLa and Ca Ski cervical cancer cell lines, with a favorable selectivity index compared to red blood cells and fibroblasts.
