rWTC-MBTA is a **personalized, autologous whole-cell cancer vaccine** formulated to stimulate adaptive and innate immune responses against solid tumors, including glioblastoma multiforme and other immunologically "cold" cancers[4][1][2][7][9][10]. The vaccine comprises irradiated whole tumor cells (rWTC) from the patient, coated with Mannan-BAM, and further combined with Toll-like receptor agonists—lipoteichoic acid (TLR2 agonist), polyinosinic:polycytidylic acid (TLR3 agonist), and resiquimod (TLR7/8 agonist)—plus an anti-CD40 monoclonal antibody[4][7]. This combination activates dendritic cells and both arms of the immune system: mannan triggers phagocytosis via pattern recognition receptors; TLR agonists induce pro-inflammatory and antigen-presenting activities; and anti-CD40 mAb delivers costimulatory signals, ensuring robust dendritic cell and T-cell activation. In preclinical studies, rWTC-MBTA led to complete tumor regression and immune memory in murine glioblastoma and other solid tumor models, preventing tumor recurrence upon rechallenge[2][1][4]. The vaccine is currently in preclinical and early translational stages for glioblastoma, triple-negative breast cancer, and colon cancer, and is designed to be produced using patient-derived tumor cells post-surgical resection[2][4][7][10].
