Overview

**RX-3117 + nab-paclitaxel** is a combination regimen being investigated as a first-line treatment for metastatic pancreatic cancer. RX-3117 is an orally administered small molecule antimetabolite nucleoside analog, selectively activated by uridine cytidine kinase 2 (UCK2), which is overexpressed in cancer cells. Once activated, RX-3117 is incorporated into cellular DNA and RNA, inhibiting their synthesis and triggering apoptotic cell death in tumors. nab-paclitaxel (brand name: Abraxane) is an albumin-bound formulation of paclitaxel, a classic microtubule inhibitor used in various solid tumors. The combination has demonstrated promising tolerability, safety, and early efficacy in phase I/II trials in newly diagnosed metastatic pancreatic cancer, with overall response rates around 23.1% and a disease control rate of 74.4%[1][2][3][5][6]. RX-3117 is particularly notable for its oral administration and potentially improved safety and pharmacokinetics over similar agents such as gemcitabine.
