**RXC004 + pembrolizumab** is an investigative combination therapy comprising RXC004 (zamaporvint), an oral small molecule inhibitor of Porcupine in the Wnt signaling pathway, and pembrolizumab, a monoclonal antibody PD-1 checkpoint inhibitor. RXC004 is designed to inhibit Wnt-ligand dependent tumor growth and modulate immune evasion, potentially sensitizing tumors to immune checkpoint blockade. This combination is being evaluated in Phase 2 clinical trials for genetically selected solid tumors, primarily advanced biliary tract cancer, microsatellite stable metastatic colorectal cancer (MSS mCRC), and RNF43-mutant pancreatic cancer.
