Overview

**RYZ401** is a novel, investigational radiopharmaceutical therapy designed for the treatment of solid tumors expressing somatostatin receptors (SSTRs)[1][7]. It is labeled with Actinium-225 (Ac-225), a potent alpha-emitting radioisotope intended to deliver targeted cytotoxicity by binding the somatostatin receptor on tumor cells, resulting in localized DNA damage and cell death. The mechanism involves selective targeting of SSTR-expressing tumor cells, leveraging the high linear energy transfer of alpha particles to achieve enhanced anti-tumor efficacy. The primary indication under investigation includes neuroendocrine tumors and other SSTR-positive solid tumors. The developer is RayzeBio; Phase 1 clinical trials are planned as of mid-2025[1][7].
