Overview

RZ-629 is a novel, orally administered, gut-restricted small-molecule G protein-coupled receptor 40 (GPR40) agonist developed by Rezubio Pharmaceuticals. It utilizes a membrane-anchored drug (MADD) design to restrict its activity to the gut, thereby minimizing systemic exposure and reducing the risk of liver and pancreas toxicities. Preclinical studies have demonstrated that RZ-629 improves glucose control and reduces body weight in animal models, with nearly all of the parent compound excreted in feces and minimal presence in bile or urine. The drug is being developed primarily for diabetes mellitus, with additional potential for metabolic diseases such as obesity[1][2][3][6].
