Overview

RZ-iNK001 is an allogeneic, induced pluripotent stem cell (iPSC)-derived natural killer (NK) cell therapy engineered with a chimeric antigen receptor (CAR) targeting an undisclosed solid-tumor antigen. Developed by Guangzhou Regenverse Therapeutics, the therapy utilizes multiplex gene editing to enhance antitumor activity and overcome the immunosuppressive tumor microenvironment. Specifically, it incorporates NKG2D and CD16a to boost recognition and cytotoxicity, and CXCR1/2 to improve trafficking to solid tumors. Additionally, the cells feature knockouts (KO) of CISH, TGFβR2, and CD38 to enhance persistence, metabolic fitness, and resistance to TGF-β-mediated suppression and fratricide. RZ-iNK001 is currently in preclinical development for the treatment of solid tumors, including lung cancer and gliomas.
