S-(-)-FPFS-1181 is a synthetic small molecule and an analogue of the neuroprotective agent R-(-)-BPAP (1-(benzofuran-2-yl)-2-propylaminopentane). It is chemically identified as the S-(-) enantiomer of N-(2-propynyl)-1-(benzofuran-2-yl)-2-propylaminopentane. Developed by Fujimoto Pharmaceutical, it belongs to the catecholaminergic and serotonergic activity enhancer (CAE/SAE) class of compounds, which are designed to improve symptoms of neurodegenerative disorders by increasing the impulse-evoked release of monoamine neurotransmitters. In preclinical studies using human dopaminergic SH-SY5Y cells, S-(-)-FPFS-1181 was evaluated for its ability to prevent apoptosis induced by the neurotoxin N-methyl(R)salsolinol. However, unlike its parent compound and certain other analogues, S-(-)-FPFS-1181 failed to stabilize mitochondrial membrane potential or induce the expression of the anti-apoptotic protein Bcl-2, indicating that its neuroprotective efficacy is significantly lower than that of its R-enantiomer counterparts.
