Overview

S-(2-oxo)pentadecylcoa is a synthetic small molecule derivative of coenzyme A featuring a long-chain fatty acid (specifically a 2-oxopentadecyl group) attached via a thioester bond. It is classified as an S-alkyl-CoA and functions primarily as an experimental biochemical tool. This compound acts as a nonhydrolyzable analog of myristoyl-CoA and serves as a potent inhibitor of myristoyl-CoA:protein N-myristoyltransferase (NMT), the enzyme responsible for transferring myristoyl groups to the N-terminal glycine residue of target proteins[3][5]. By inhibiting NMT, it disrupts protein myristoylation—a modification essential for membrane localization and function of various signaling proteins. Its main applications are in biochemical research on lipid metabolism, protein modification pathways (notably myristoylation), and antifungal drug development targeting NMT[3][9]. There are no approved therapeutic indications or clinical trials for this compound.
