S-1 + bevacizumab + cisplatin is a combination chemotherapy regimen used primarily in the treatment of advanced non-squamous non-small cell lung cancer (NSCLC). S-1 is an oral fluoropyrimidine derivative that acts as a prodrug of 5-fluorouracil (5-FU), inhibiting thymidylate synthase and thereby interfering with DNA synthesis. Cisplatin is a platinum-based chemotherapeutic agent that forms DNA crosslinks, leading to apoptosis. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis required for tumor growth. This combination has demonstrated high response rates and acceptable toxicity profiles in phase II clinical trials for chemotherapy-naïve patients with advanced NSCLC[1][2][3].
