S-1 + nedaplatin is a combination chemotherapy regimen consisting of two agents: - **S-1** is an oral fluoropyrimidine derivative composed of tegafur (a prodrug of 5-fluorouracil), gimeracil (an inhibitor of dihydropyrimidine dehydrogenase to increase 5-FU bioavailability), and oteracil potassium (to reduce gastrointestinal toxicity). This combination has been studied for the treatment of various squamous cell carcinomas, including head and neck squamous cell carcinoma (HNSCC), non-small cell lung cancer (NSCLC) with squamous histology, and nasopharyngeal carcinoma. The typical administration involves oral S-1 for 14 days combined with intravenous nedaplatin on day 1 or day 8 in a three-week cycle[2][3][4][5]. The regimen aims to provide effective cytotoxic therapy with improved tolerability over traditional platinum/fluoropyrimidine combinations.
