S-1226 is a novel inhaled therapeutic composed of a mixture of carbon dioxide-enriched air and nebulized perflubron. It is being developed as a bronchodilator for respiratory diseases such as asthma, cystic fibrosis, and COVID-19 by SolAeroMed. Unlike traditional bronchodilators that act on β2-adrenergic or muscarinic receptors, S-1226 exerts its effects through both biophysical and pharmacological mechanisms. The carbon dioxide component provides rapid bronchodilation by relaxing airway smooth muscle, while perflubron acts as a synthetic surfactant with mucolytic properties to help clear mucus and reduce inflammation. This dual action allows for immediate relief of airway constriction and improved mucus clearance without the receptor-mediated tolerance seen in conventional therapies[1][3][4][5][8]. Clinical studies have shown that S-1226 is safe and well tolerated in both healthy subjects and patients with respiratory diseases[4][5]. It has demonstrated efficacy in rapidly dilating airways during acute asthma exacerbations and improving oxygenation in cystic fibrosis patients[4][8].
