S-234462 is a small molecule drug candidate developed as an anti-obesity agent. It acts as an insurmountable antagonist of the neuropeptide Y (NPY) Y5 receptor, a target implicated in the regulation of energy homeostasis, including stimulation of food intake and inhibition of energy expenditure. In preclinical studies, S-234462 demonstrated high affinity for the NPY Y5 receptor and exhibited superior anti-obesity effects compared to surmountable antagonists such as S-2367. The compound effectively suppressed food intake induced by selective Y5 receptor agonists and reduced body weight gain in high-fat diet-induced obese mice. However, during early clinical development, there were reports of liver impairment associated with its use, prompting further investigation into its safety profile[3][4]. The drug was developed by Shionogi as a backup to S-2367 but was ultimately discontinued in favor of follow-up compounds due to efficacy and safety considerations[2][4].
