Overview

S-237648 is an orally active small molecule inhibitor of monoacylglycerol acyltransferase 2 (MGAT2) being developed by Shionogi for the treatment of obesity and associated metabolic conditions. MGAT2 is an enzyme primarily expressed in the small intestine that plays a critical role in the re-synthesis of triglycerides from dietary monoacylglycerol and fatty acids. By inhibiting MGAT2, S-237648 aims to decrease the efficiency of fat absorption and potentially modulate the release of gut hormones such as glucagon-like peptide-1 (GLP-1) and peptide YY (PYY), which are involved in appetite regulation and glucose homeostasis. The drug is currently in Phase 2 clinical trials evaluating its metabolic effects in Japanese patients with obesity, visceral adiposity, impaired glucose tolerance, type 2 diabetes, and hyperlipidemia.
