S-309309 is a novel, orally administered small molecule inhibitor of monoacylglycerol O-acyltransferase 2 (MGAT2), developed primarily for the treatment of obesity. Unlike GLP-1 receptor agonists, S-309309 targets MGAT2, an enzyme involved in triglyceride synthesis and fat absorption in the intestine. Preclinical studies have shown that S-309309 enhances intestinal beta oxidation, increases energy expenditure, suppresses food intake and body weight gain, improves insulin sensitivity, and reduces fatty liver in diet-induced obese mice. Clinical trials have demonstrated acceptable safety and tolerability profiles with no serious adverse events or significant cardiac effects observed. The drug is currently being evaluated in Phase II clinical trials to assess its efficacy on body weight reduction in adults with obesity[1][3][4][5].
