Overview
S-524101 is a next-generation sublingual immunotherapy (SLIT) tablet developed by Shionogi for the treatment of perennial allergic rhinitis induced by house dust mites (HDM). It is a new formulation of the HDM allergen extract previously marketed by Shionogi as Actair (S-524111), designed to improve patient convenience. The drug consists of standardized extracts from the mites *Dermatophagoides farinae* and *Dermatophagoides pteronyssinus*. S-524101 works through allergen-specific immunotherapy, which aims to modify the underlying allergic disease by inducing immunological tolerance. This process involves the induction of regulatory T cells (Tregs), a shift from a Th2 to a Th1 immune response, and the production of allergen-specific IgG4 antibodies that block IgE-mediated mast cell degranulation. The product is currently in Phase 3 clinical development in Japan.
