S 64315 + azacitidine is a combination therapy under investigation for the treatment of acute myeloid leukemia (AML). S 64315 (also known as MIK665 or MIK-665) is a potent, selective small molecule inhibitor of myeloid cell leukemia 1 (Mcl-1), an anti-apoptotic protein in the Bcl-2 family that is often upregulated in cancer cells and promotes tumor cell survival. By inhibiting Mcl-1, S 64315 induces apoptosis in malignant cells. Azacitidine is a hypomethylating agent that incorporates into DNA and RNA, leading to cytotoxicity and reactivation of silenced genes, which contributes to its antineoplastic effects. The combination aims to enhance pro-apoptotic activity against AML by targeting complementary pathways involved in leukemic cell survival[2][6][8].
