S-8117 is a tamper-resistant, controlled-release (CR) oral formulation of **oxycodone hydrochloride** developed by **Shionogi** for the management of chronic pain. As a semi-synthetic opioid, oxycodone acts primarily as an agonist at the **mu-opioid receptor**, providing potent analgesia by modulating pain perception in the central nervous system. The S-8117 formulation incorporates specific tamper-resistant (OTR) features designed to prevent common methods of abuse, such as crushing, dissolving, or chewing the tablets for rapid drug release. Shionogi has advanced S-8117 into Phase 3 clinical development in Japan, specifically targeting chronic non-cancer pain conditions such as chronic low back pain, osteoarthritis, diabetic neuropathic pain, and postherpetic neuralgia. Clinical studies have focused on evaluating the safety and efficacy of switching patients from conventional oxycodone CR formulations to the S-8117 tamper-resistant version.
