Overview

S 81694 is a potent and selective small molecule inhibitor of monopolar spindle 1 kinase (MPS1), also known as TTK protein kinase. MPS1 is a serine/threonine kinase involved in the regulation of the mitotic checkpoint during cell division. By inhibiting MPS1/TTK activity, S 81694 disrupts proper chromosome segregation and leads to cell death in rapidly dividing cancer cells. The drug was developed for use as an antineoplastic agent in patients with advanced or metastatic solid tumors and breast cancer who had exhausted standard treatment options. Clinical studies included both monotherapy and combination therapy with paclitaxel. Development was discontinued after early-phase trials due to limited efficacy compared to emerging therapies and a narrow therapeutic window[1][2][5][6].
