S-8484 is a novel engineered immunoglobulin E (IgE) protease therapeutic developed by Seismic Therapeutic. It is designed to selectively cleave IgE antibodies, both in circulation and on the surface of B cells, thereby targeting multiple mechanisms of humoral immune dysfunction in allergic diseases. The drug leverages Seismic’s IMPACT platform for enhanced selectivity and drug-like properties, aiming to induce rapid and sustained reduction of IgE levels. Preclinical studies have demonstrated potent, rapid, and highly selective cleavage of soluble IgE as well as concentration-dependent cleavage of the IgE B cell receptor and reduction in high-affinity FcεRI expression on mast cells. In animal models, S-8484 showed efficacy comparable to approved anti-IgE therapies for conditions such as anaphylaxis and allergy. The primary indications under investigation include food allergies, chronic rhinosinusitis with nasal polyps, chronic spontaneous urticaria, and other allergic diseases[1][2][6].
