S-892216-LAI is a long-acting injectable formulation of S-892216, a second-generation small molecule antiviral developed for pre-exposure prophylaxis and treatment of COVID-19. S-892216 is a potent and selective inhibitor of the SARS-CoV-2 main protease (3C-like protease, 3CL^pro^), an essential enzyme for viral replication. Preclinical studies demonstrate strong enzyme inhibitory and antiviral activity against multiple SARS-CoV-2 variants, including Omicron, with efficacy at lower doses than currently approved 3CL^pro^ inhibitors such as nirmatrelvir and ensitrelvir. The LAI formulation utilizes a wet-milled nanosuspension to achieve a sustained-release profile for extended pharmacokinetic exposure.
